In theory yes if you check the option "keep offline" on controller settings, save and then sync. Once you compiled the rules and devices succesfully (right side of the API sync button) your controller should keep running the rules even without internet connection. Any rule that depends on virtual weather stations or uses the cloud will not work and/or act funny for obvious reasons.
If you don't check "keep offline" option your controller will keep rebooting until internet is restored, to be honest I have never understood why would someone would like to have their controller rebooting if there is no internet.
